Effects of nitrogen and potassium on the chemical composition of coffee beans and on beverage quality Agronomy
Clemente, Junia Maria; Martinez, Herminia Emilia Prieto; Alves, Leonardo Corrêa; Finger, Fernando Luíz; Cecon, Paulo Roberto.
The ratio of nitrogen (N) to potassium (K) is important in the production of specialty coffees because the relative amounts of N and K can either suppress the formation or increase the concentration of compounds that are essential to the flavor and aroma of specialty coffees. The purpose of this study was to evaluate the effect of different N:K ratios (w/w) and K doses on the cup quality of coffee. The concentrations of chemical compounds essentials to achieve good flavor and aroma, the N and K contents, and the caffeine contents of coffee leaves and beans were evaluated in this study. The N:K ratio and the K dose were found to be important factors in cup quality, the best quality corresponding to an N:K ratio of 1:1.56. The best cup quality was obtained...

Genetic investigations on the caffeine and chlorogenic acid relationship in an interspecific cross between Coffea liberica dewevrei and C-pseudozanguebariae ArchiMer
Ky, Chin-long; Barre, Philippe; Noirot, Michel.
The objective of the paper was to identify the number of major loci explaining caffeine content in coffee seeds. Investigations were based on previously published results: (1) Caffeine binds to chlorogenic acids in a 1:1 molecular ratio; (2) Between species, the caffeine content is correlated to the chlorogenic acid content; (3) Only a part of chlorogenic acids is bound to caffeine. Especially, the content ratio between caffeine and chlorogenic acids varied between species. For identifying the number of major loci, a quantitative trait locus (QTL) approach was carried out using an interspecific cross between two highly differentiated species-Coffea liberica dewevrei and Coffea pseudozanguebariae, the latter being a caffeine-free species. As main finding,...

Hydroxypropylcellulose controlled release tablet matrix prepared by wet granulation: effect of powder properties and polymer composition BABT
Teixeira,Antonio Zenon Antunes.
The aim of this study was to attain 100% drug release of caffeine after 24 h from hydroxypropylcellulose (HPC) tablet matrices and to investigate the effect of co-excipient. Physical properties of the powders were evaluated and suggested for a wet granulation process. The tablet containing caffeine was formulated by different weight ratios of hydrophilic polymers. The results of polymer evaluation confirmed that the increase of HPC level with the same drug content significantly decreased the rate of drug release. The presence of co-polymer excipients carboxymethylcellulose (CMC) and polyvinylpyrrolidone (PVP) in the tablet matrix was also investigated. The release rate was also controlled by low levels of CMC (<10%) while PVP did not show any...

Inhibition of the sarcoplasmic reticulum Ca2+ pump with thapsigargin to estimate the contribution of Na+-Ca2+ exchange to ventricular myocyte relaxation BJMBR
Bassani,R.A.; Bassani,J.W.M..
Relaxation in the mammalian ventricle is initiated by Ca2+ removal from the cytosol, which is performed by three main transport systems: sarcoplasmic reticulum Ca2+-ATPase (SR-A), Na+-Ca2+ exchanger (NCX) and the so-called slow mechanisms (sarcolemmal Ca2+-ATPase and mitochondrial Ca2+ uptake). To estimate the relative contribution of each system to twitch relaxation, SR Ca2+ accumulation must be selectively inhibited, usually by the application of high caffeine concentrations. However, caffeine has been reported to often cause changes in membrane potential due to NCX-generated inward current, which compromises the reliability of its use. In the present study, we estimated integrated Ca2+ fluxes carried by SR-A, NCX and slow mechanisms during twitch...

Metabolism of alkaloids in coffee plants Braz. J. Plant Physiol.
Ashihara,Hiroshi.
Coffee beans contain two types of alkaloids, caffeine and trigonelline, as major components. This review describes the distribution and metabolism of these compounds. Caffeine is synthesised from xanthosine derived from purine nucleotides. The major biosynthetic route is xanthosine -> 7-methylxanthosine -> 7-methylxanthine -> theobromine -> caffeine. Degradation activity of caffeine in coffee plants is very low, but catabolism of theophylline is always present. Theophylline is converted to xanthine, and then enters the conventional purine degradation pathway. A recent development in caffeine research is the successful cloning of genes of N-methyltransferases and characterization of recombinant proteins of these genes. Possible biotechnological...

Mineral nutrition and caffeine content in coffee leaves Bragantia
MAZZAFERA,PAULO.
The effect of nutrient supply on the caffeine content of coffee (Coffea arabica L.) leaves was investigated. Seeds were germinated in nutrient-agar media lacking N, P, K, Ca, Mg, S, Zn, B or Mo. The control treatment contained all essential nutrients. The caffeine concentration was determined seven months after seed sowing when the seedlings have 3 to 4 pair of leaves. The omission of K induced the highest caffeine content in the leaves (24.5 g.kg-1). Caffeine in the control treatment was 21.9 g.kg -1. Absence of P induced the lowest content, 17.5 g.kg-1.

Modulation by caffeine of calcium-release microdomains in frog skeletal muscle fibers Biol. Res.
VERGARA,JULIO L; DIFRANCO,MARINO.
The effects of caffeine on the process of excitation-contraction coupling in amphibian skeletal muscle fibers were investigated using the confocal spot detection technique. This method permits to carefully discriminate between caffeine effects on the primary sources of Ca2+ release at the Z-lines where the triads are located and secondary actions on other potential Ca Release sources. Our results demonstrate that 0.5 mM caffeine potentiates and prolongs localized action-potential evoked Ca2+ transients recorded at the level of the Z-lines, but that 1mM only prolongs them. The effects at both doses are reversible. At the level of the M-line, localized Ca2+ transients displayed more variability in the presence of 1 mM caffeine than in control conditions. At...

The Ginoide Hydrolipodystrophy (GHLD), commonly known as cellulite, occurs in 80-90% of the female population after the puberty period and comes from a metabolic modification in the cutaneous adipose tissue. Caffeine has been used in topical formulations due to its lipolytic action. We studied a nanoemulsion (F3) containing caffeine with two surfactants (oleth-3 and oleth-20) by emulsification method by phase inversion temperature inversion (PIT). The polydispersion indices (PDI) showed the reduced deviation of 0.1. The mean droplet size was ~ 40 nm. The evaluated constant of Ostwald, in the refrigerator condition was the most favorable during the stability test. In the In Raman spectroscopy assay, the caffeine bands found in F3 were compatible with those...

Kasabova-Angelova,Alexandra; Kondeva-Burdina,Magdalena; Mitkov,Javor; Georgieva,Maya; Tzankova,Virginia; Zlatkov,Alexander.
The effects of new derivatives of caffeine-8-thioglycolic acid (100 µM) on isolated rat brain synaptosomes, human neuroblastoma cell line SH-SY5Y and human recombinant MAOB enzyme (hMAOB) (1 µM) were evaluated. Most of the compounds, administered alone, didn’t show statistically significant neurotoxic effects on SH-SY5Y, when compared to the control (non-treated cells). Of all studied structures JTA-2Ox, JTA-11, JTA-12 and JTA-13 decreased cell viability. In combination with 6-hydroxydopamine (6-OHDA) (100 µM), only JTA-1 and JTA-2 revealed neuroprotective effects, stronger than those of caffeine. All compounds administered alone revealed, neurotoxic effects on synaptosomes, as compared to non-treated synaptosomes. JTA-1, JTA-2 and JTA-3 showed lowest...

Phenolic compounds, methylxanthines and antioxidant activity in cocoa mass and chocolates produced from "witch broom disease" resistant and non resistant cocoa cultivars Ciência e Agrotecnologia
Leite,Paula Bacelar; Maciel,Leonardo Fonseca; Opretzka,Luiza Carolina França; Soares,Sergio Eduardo; Bispo,Eliete da Silva.
The "witch broom disease" caused by the fungus called Moniliophthora perniciosa is one of the most important cocoa diseases in Latin America, causing around 70% production reduction in the southern Bahia. In attempt to solve the problem, many cultivars resistant to the disease have been recommended to farmers. On the other hand, the chocolate flavour is composed by many compounds whose formation depends on the genetic background, environment where cocoa is grown and processing operations. Therefore, this work aimed at determining the monomeric phenolic compounds, methylxanthines and antioxidant activity of cocoa mass and dark chocolate from cocoa cultivars resistant to "witch broom disease" and non resistant to the disease. The total phenolic compounds in...

Ploidy level and caffeine content in leaves of Coffea Scientia Agricola
Silvarolla,Maria Bernadete; Mazzafera,Paulo; Lima,Marinez Muraro Alves de; Medina Filho,Herculano Penna; Fazuoli,Luiz Carlos.
Several species of Coffea and varieties of C. arabica differing in chromosome number had their caffeine content determined in the leaves. For Coffea arabica var. "Angustifolia", var. "Caturra" and var. "Icatu", it was observed caffeine decrease from the haploid (2n = 22) to tetraploid leaves (2n = 44). Caffeine in the tetraploid "Angustifolia" was decreased by 50% when compared to the haploid. Caffeine reduction was also observed in leaves of C. canephora var. "Kouilou" as chromosome number was increased (2n = 22 ® 2n = 44). In this case, caffeine in the leaves of the diploid genotype was close to 4 times higher than in the tetraploid. On the other hand it was observed an increase of the alkaloid when the chromosome number was doubled in the C. canephora...

Roles of nibrin and ATM/ATR kinases on the G2 checkpoint under endogenous or radio-induced DNA damage Biol. Res.
Marcelain,Katherine; de la Torre,Consuelo; González,Patricio; Pincheira,Juana.
Checkpoint response to DNA damage involves the activation of DNA repair and G2 lengthening subpathways. The roles of nibrin (NBS1) and the ATM/ATR kinases in the G2 DNA damage checkpoint, evoked by endogenous and radio-induced DNA damage, were analyzed in control, A-T and NBS lymphoblast cell lines. Short-term responses to G2 treatments were evaluated by recording changes in the yield of chromosomal aberrations in the ensuing mitosis, due to G2 checkpoint adaptation, and also in the duration of G2 itself. The role of ATM/ATR in the G2 checkpoint pathway repairing chromosomal aberrations was unveiled by caffeine inhibition of both kinases in G2. In the control cell lines, nibrin and ATM cooperated to provide optimum G2 repair for endogenous DNA damage. In...

Sobre a Padronização de um Método Prático Para a Dosagem da Cafeína BABT
Bührer,Nilton E..
In this work the author puts in evidence a practical and at the same time rapid method for the determination of the cafein quantity in erva-mate, that may be utilized to perfection in food and industrial analyses or others that not demand scientific rigour. The author presents a description of the method and makes a comparison with the classical method of Grandval-Lajoux, showing that the differences between the one and the other are practically nil. In conclusion, the result of the three analyses of the same sample of erva-mate, made by the Grandval-Lajoux method are cited, show that the same differences appear as those shown between the rapid method suggested by the author and the classical method. In this case, the maximum difference was 0,06% and the...
